Recipe for S2 Cell Nucleofection Solution

We use this solution instead of a commercial nucleofection kit to transfect cultured Drosophila S2 cells when using a commercial electroporator, program G30.

Concentrations below are final concentrations.

• 100mM Na2HPO4
• 20mM NaH2PO4
• 5mM KCl
• 15mM MgCl2
• 50mM D-mannitol
• pH7.2 (some minor adjustment may be needed to reach 7.2)

Sterile filter solution and aliquot.
Storage at 4C is sufficient to keep the nucleofection solution good for extended periods.
